a Microsatellite locus name and primer sequence after Lu et al., 2001; b From Lu et al., 2001;c This study; d These data are based on results from 15 wild-born giant pandas. The unlabelled primers for all microsatellites except AME-|28A and AME-m 28B have the sequence GTGTCTT attached to the 5' end; thus, the PCR products are larger than previously reported (Lu et al., 2001).

Tian (SB 458), the male giant panda on loan to the Smithsonian's National Zoological Park in Washington, DC. Tian Tian's dam is Yong Ba (SB 397). His sire is uncertain because Yong Ba was naturally mated to Pan Pan (SB 308) and artificially inseminated with semen from Lin Nan (SB 298) and Lo Lo (SB 305). Microsatellite markers are inherited in a Mendelian fashion so, barring a mutation, Tian Tian's genotype must be derived from a combination of alleles present in Yong Ba and his sire. Lin Nan is excluded as the sire because he does not share any allele with Tian Tian at Ame-^13, Ame-^27 and Ame-^70. (This failure of allele sharing between an offspring and a possible sire is termed an Offspring [Off]-Sire loci mismatch in Table 10.3, column 12; see below.) Lin Nan

a Yong Ba (SB 397) is the known dam of Tian Tian (SB 458). Yong Ba was naturally mated to SB 308 and artificially inseminated with semen from SB 298 and SB 305 so the paternity of Tian Tian is uncertain. Both SB 298 and SB 305 are excluded as potential sires at multiple loci. Tian Tian resulted from the natural mating with Pan Pan (SB 308). h NM, natural mating.c Al, artificial insemination. d Bold, Excluded as sire. e , Excluded with selected dam Yong Ba (SB 458).1O, no product detected.
