Osteogenic Differentiation in the HACollagen Hybrid

When the MSCs were stimulated by ODM for 5 days, the intensity of ALP staining was greater in the HA-collagen hybrid gel than in the collagen mono gel (Figure 1A). The ALP activity was also significantly two times greater in the HA-collagen hybrid gel than in the collagen mono gel (Figure 1B).

Alizarin red staining for the MSC gel culture showed enhanced calcium deposition in the extracellular regions of the gel cultures maintained in ODM. The intensity of the Alizarin red staining on day 10 was greater in the HA-collagen hybrid gel than in the collagen mono gel (Figure 2A). Furthermore, the incorporation of calcium into the extracellular matrix on day 14 was also significantly greater in the HA-collagen hybrid gel than in the collagen mono gel (Figure 2B).

The gene expressions of all bone markers examined, BSP, ALP, type I collagen, Runx2 and osterix were significantly greater in the HA-collagen hybrid gel than in the collagen mono gel (Figure 3). The relative increase in gene expression levels for BSP, ALP, type I collagen, Runx2 and osterix were 1.5-fold, 1.8-fold, 1.2-fold, 1.3-fold and 1.7-fold, respectively, as compared to those in the collagen mono gel.
