3.1. Characterization of MSCs

In human adult bone marrow, stem cell population is mainly composed of haematopoietic stem cells (HSC) and a very small non-HSC portion consists of MSCs. CD34 is a surface marker for HSC and CD44 is one of the MSC markers. In the present study, over 95% in vitro expansed cells were CD34-/CD44+. MSC cytogenetic analysis showed normal diploid karyotype (46XY) and normal chromosome structure, which suggest that genetic stability was retained prior to autotransplantation.

3.2. The Changes of Heart Morphology and Cardiac Function

After the MSC transplantation, the internal diameters of all atria and ventricles became smaller compared with pre-transplantation (Table 1). Further changes were observed during follow up on different time points up to six months. Similar measurements were also taken for the diameters of aorta and pulmonary artery as well as the thicknesses of septum and posterior wall of left ventricle and no difference was detected between pre- and posttransplantation.

As shown in Table 2, all assessed cardiac function parameters were improved after MSC administration and showed the best performance at six months (6 m). Stroke volume (SV), fractional shortening (FS) and left ventricular ejection fraction (LVEF) were increased by 20%, 52% and 29% respectively after 6 months.

3.3. The Improvement of Myocardial Perfusion

Figure 1 showed the myocardial perfusion status before cell therapy and the changes after MSC administration. The most apparent improvement was observed at one month (1m) especially in the YZ dimension.
