Temperature is not the only parameter affecting protein-state transition. Other components in solution may have an equally profound effect. Figure 11.8 shows the effect of urea concentration in the solution containing the protein on the intensity of the fluorescence of the solution stimulated by the light of the appropriate wavelength. The straight-line portions at either end of the curve represent fluorescence of 100% folded state at low urea concentrations and 100% unfolded state at high urea concentrations. Increasing urea concentration has the same effect on protein stability as increasing temperature.
